The biosynthetic route from ornithine to proline.

چکیده
It is shown by tracer experiments with DL-[2-3H,5-14C]- and DL-[(RS)-5-3H,5-14C]rnithine, that the metabolic conversion of ornithine into proline, in three plant species (Nicotiana tabacum, Datura stramonium, and Lupinus angustifolius), takes place with maintenance of the delta-hydrogen atoms but with loss of the alpha-hydrogen atom. This indicates a route via alpha-keto-delta-aminovaleric acid (5-amino-2-oxopentanoic acid) and disproves the accepted route via glutamic gamma-semialdehyde (2-amino-5-oxopentanoic acid).

Ornithine cyclodeaminase-based proline production by Corynebacterium glutamicum
BACKGROUND The soil bacterium Corynebacterium glutamicum, best known for its glutamate producing ability, is suitable as a producer of a variety of bioproducts. Glutamate is the precursor of the amino acid proline. Proline biosynthesis typically involves three enzymes and a spontaneous cyclisation reaction. Alternatively, proline can be synthesised from ornithine, an intermediate of arginine bi...
متن کاملMethanococcus jannaschii generates L-proline by cyclization of L-ornithine.
Cell extracts of Methanococcus jannaschii have been shown to readily convert L-ornithine to L-proline. This cyclization reaction proceeds with the loss of only the C-2 nitrogen, as has been documented for ornithine cyclodeaminase (EC 4.3.1.12). Since no gene homologous to that coding for ornithine cyclodeaminase is present in the genome of M. jannaschii, these results indicate that proline bios...
متن کاملArginine, ornithine, and proline interconversion is dependent on small intestinal metabolism in neonatal pigs.
We have previously shown that arginine deficiency is exacerbated by the removal of dietary proline in orally, but not parenterally, fed piglets. Therefore, we hypothesized that the net interconversions of proline, ornithine, and arginine primarily occur in the small intestine of neonatal piglets. Ten intragastrically fed piglets received either intraportal (IP) or intragastric (IG) primed, cons...
متن کاملTransforming growth factor-beta(1) stimulates L-arginine transport and metabolism in vascular smooth muscle cells: role in polyamine and collagen synthesis.
BACKGROUND Transforming growth factor-beta(1) (TGF-beta(1)) contributes to arterial remodeling by stimulating vascular smooth muscle cell (VSMC) growth and collagen synthesis at sites of vascular injury. Because L-arginine is metabolized to growth-stimulatory polyamines and to the essential collagen precursor L-proline, we examined whether TGF-beta(1) regulates the transcellular transport and m...
متن کاملGlutamate, Ornithine, Arginine, Proline, and Polyamine Metabolic Interactions: The Pathway Is Regulated at the Post-Transcriptional Level
The metabolism of glutamate into ornithine, arginine, proline, and polyamines is a major network of nitrogen-metabolizing pathways in plants, which also produces intermediates like nitric oxide, and γ-aminobutyric acid (GABA) that play critical roles in plant development and stress. While the accumulations of intermediates and the products of this network depend primarily on nitrogen assimilati...
